that's exactly what happened yesteRday.. this is gonna be a Ranting entRy..
i had my 2nd LSM2201 biochem lab yesteRday afteRnoon...
we weRe supposed to do gel filtRation of ouR pRotein sample puRified paRtially fRom the pRevious pRactical.. Right fRom the staRt of the expeRiment, my gRoup(2 guys, 2 gals) was alReady at a disadvantage cos the guys who did the puRification pRevious lab session did wRongly and oveR dilute the sample.. in the end we had double the sample size of otheRs... it was only the beginning....
this week we let the guys do the pRotein standaRds cos we giRls did it last week.. so we set the sephadex gel in the column instead.. it took almost 2-3 houRs foR the stupid gel to set, and we weRe just theRe pouRing and staRing at the column, wasting time... finally when we weRe done, we set up the peRistaltic pump to Run the buffeR NaCl thRough the column... it was all fine at fiRst...
till.. the chief TA came oveR.. he took a look at the dRipping buffeR, says the Rate is too slow.. took an eppendoRf tube to test the Rate, Realised it's okay then walk away... then ouR pump failed... we panickly shut the column at the bottom and called ouR TA foR help... wasted lots of time... by this time ppl weRe alReady Running theiR pRotein samples...
ouR sample as i said was too diluted.. and ouR volume was double... while otheR ppl's sample was gReenish/yellowish, ouRs was cleaR and tRanspaRant. sianz...
set up the fRaction collectoR.. adjusted to collect 125 dRops which is appRox 3ml peR tube.. staRting Running it... muRphy's law came tRue again... the fRaction collectoR, instead of collecting 3ml peR tube, it collected bout 8ml foR the fiRst tube, 5ml foR second tube, 10ml foR thiRd tube and things like that... ha... ouR TA was shocked... so in the end, while otheR ppl collect 20 tubes, we only collect 5 tubes..
that was not all... we had to peRfoRm enzyme essay on the 5 fRactions, and accoRding to the pRotocol we had to put the tubes into a wateR bath of 37 degRees celcius foR 15mins.. we we went and put the tubes in the wateR bath, a minute lateR, a TA exclaimed why the wateR bath was 60 degRees celcius!!! we quickly took out ouR tubes but it was too late... enzymes denatuRed... aRghhhhh!!!!!! the technician came and put the sign "do not use" on the wateR bath at the moment.... too late!!!!!!! sianz........
we also kena scolding foR putting the eppendoRf tube into the micRowave... and to think we actually asked a TA last week and that TA said what we do was Right...........
having lots of pRob with the machines and equipments is bad enough... it makes it woRse when u have a hoRRible lab mate......
that iRRitating guy with nothing to do, stood by me(veRy neaR somemoRe) and staRe at what i was doing, not offeRing any help.. even when we went to thRow away the ammonium sulphate in the waste bottle, he asked what we weRe doing...... DUH?!?!!? anthony said i should have thRow him away as well.. haha..
while we weRe in the middle of an expeRiment, he came oveR and said tube 4 is
missing.. yah.. how can a sample go missing??!?!! we gals weRe absolutely suRe it was with the guys cos we passed to them eaRlieR on and they didn't RetuRn.. so this iRRitating guy just stood by my side while i was busy pipetting lots of stuff and keep chanting into my eaRs "tube 4 missing"... i told him, go and look foR it!! and he just said he can't find, then continue to stand by my side and chant.. i almost slapped him.. hah.. i so much wanted to say to him, "u think u can find it by standing heRe??!?!?!?!" but i didn't.. i can't be botheRed to talk to him le... heat was building up in me.. my paRtneR was so fRustRated that she was fRowning and just let that guy do all soRts of stupid things cos she also dunno how to stop him le.. he went to centRifuge samples that did not RequiRe centRifuging..
theRe's still lots of stupid questions and things he did that i want to Rant about... but i feel i need to cool down le.. die... dunno how to do open pRoject with this guy... these fiRst 5 lab sessions we have lab pRotocol to follow also so bad le... can't imagine open pRoject how... somemoRe this module is 6MC.. can't S/U, can't affoRd to do badly... =( this guy actually went ahead to make stupid changes in ouR lab RepoRt afteR we had alReady finalised.. and he only told us he made changed afteR the RepoRt is submitted.......... by the way, this guy has a weiRd way of doing lab RepoRt, adding lots of info that's not RequiRed, yet not answeRing questions that weRe RequiRed... how idiotic........
and the woRse thing is this... i'm in the same pRoject gRoup as him foR lsm2104 intRo to bioinfoRmatics and biocomputing.... he is the fRiend of yet anotheR iRRitating fRiend... gosh... biRds of the same featheRs flock togetheR... of couRse that's excluding me and kaiyan.. haha..
