i thought i was going to fail the gRam staining test today... =x
was totally not pRepaRed!!!
not to mention that i have
totally foRgotten the theoRy behind it (even though i eveR wRote an essay on it duRing my uni time).. all i RemembeR aRe the steps to stain... but which wall and what's the Reactions involved? i have no idea... =x
i can't even RemembeR whetheR Red is foR gRam neg oR pos!?!?
and i somehow have totally no ability to diffeRentiate cocci fRom Rods undeR the micRoscope.. it's too small!! Round looks ovalish, Oval looks Roundish.. =x
i think i was tested on this..

it looks like this undeR 100x... so SMALL!!! cocci? Rods? it somehow looks like diplococci (2 spheRical stuck togetheR) to me... but well, this is Rods.. and if i'm not wRong, it's coccobacilli... (looks halfway between spheRical and oval)
halfway thRough my STAT duty, the micRo staff came to me and sus (we weRe the only 2 left who hasn't gone foR the "tRaining")... so we got anotheR staff to coveR us while we went to do gRam stain and slide Reading..
and so, i Really heaved a big sigh of Relief when i answeRed 3 out of 4 slides Right.. haha.. the 1 slide, i was half Right and half wRong.. theRe weRe 2 oRganisms.. i identified one coRRectly and the otheR wRongly.. cos what i saw was Red cocci, but the micRo staff says it's gRam pos.. and explains it looks Red becos my decolouRising technique was wRong... =x

The decolorization step is critical and must be timed correctly; the crystal violet stain will be removed from both Gram-positive and negative cells if the decolorizing agent is left on too long (a matter of seconds).
oh well, that explains why my gRam pos bacteRia become gRam neg in my slide.. =xhey! i have neveR done gRam staining in school eveR (all done foR us, we just look into the micRoscope in school!).. and neveR had foRmal tRaining foR it at woRk... theRe was just a little piece of papeR with instRuctions stuck by the fume hood.. so that's how i leaRnt... but well.. little did i know how one holds the slide while washing can affect the staining Results! =p
it's a fRuitful lesson today.. =)
